Obiettivi formativi

The integrated course is aimed at providing students with the knowledge necessary to understand how the structure of biomolecules (mainly proteins and nucleic acids) is determined by spectroscopic and diffractometric methods, knowing how to identify which method(s) to use according to the characteristics of the system under study, and critically interpret the three-dimensional structure.

In this module (A) the basis of optical (vibrational and electronic) spectroscopies are treated in some detail and their applications in the biological field described.

Modalità di esame.

Traditionally the exam  consists of a written test and an oral test. The written test includes the answer to questions (both multiple choice and open) concerning biomolecular and X-ray NMR spectroscopy. The oral exam focus on the part of IR vibrational spectroscopy and electronic spectroscopy.

A Module: Vibrational and electronic spectroscopy (2 CFU).

This part will deal with the principles of IR vibrational spectroscopy and electronic absorption and emission (in steady state and resolved over time) and the applications of these methodologies to the study of biomolecules in different conditions (solid state, solution). In particular, the possibility of using IR spectroscopy to obtain information on the secondary and tertiary structure of proteins and the secondary structure of nucleic acids and to monitor their possible change due to interactions and / or chemical and physical modifications of the surroundings will be discussed. As for electronic spectroscopy, the aspects of quantitative sensitivity, the main types of signals sensitive to the structural organization of bio-macromolecules and metal centers possibly present, and the possibility of obtaining information on the structure and its possible modifications through the evolution of the characteristics of fluorescence signals are illustrated.

Traditionally, the course consists of frontal lectures on the blackboard with the projection of slides to support the topics covered.
